 Working Principles

Fermentation and formulation of water based flowableBacillus thuringiensis in 100L fermenter Culture stock in slant (store at 4°C) Inoculate Bt culture in 10 litre media and incubate at 30°C for 48hrs (seed culture) Fill fermentor with 100 litre of medium and sterilize (cool medium at 30°C) Inoculate seed culture and run fermentor for 5 days (Rpm-350, temperature 30±0.2°C, PH-6.8±0.2, DO-20%) (Fermentation is complete when the broth contain 90% free spores and crystals and 10% vegetative cells) Cool the broth to 5-10°C and adjust pH to 4.1±0.2 using 5M H2S Centrifuge (5000rpm for 15 min) broth and separate solid part (active ingredient)

 Applications

For the mass culture of bacteria / fungi especially Pseudomonas, Bacillus and Trichoderma.

 User Instructions

Working volume 60litres, use of 10% mother inoculum, sterilize medium by steam, allow to cool, inoculate with mother inoculum, set RPM and aeration, use antifoam agent. Run the equipment till full growth is attained.

 Working Principles

Gel electrophoresis can be used to effectively separate nucleic acids based on their size within agarose or polyacrylamide gels. Size ladders containing a range of different-length DNA fragments are run with the sample of interest to identify the corresponding size bands in a gel. Agarose gels are ideal for the separation of DNA restriction digests, PCR products, and genomic DNA or RNA prior to Southern or northern blotting. Polyacrylamide gels can be used for what is known as polyacrylamide gel electrophoresis, or PAGE. Gels can be hand cast in the lab, or precast gels can be purchased from commercial provider

 Applications

Applications include oligonucleotide analysis, RNase protection assays, and northern blotting. Non-denaturing polyacrylamide gels are used to separate small double-stranded DNA (dsDNA) fragments, particularly PCR products. Denaturing polyacrylamide gels are used for separation of small RNA and single-stranded DNA (ssDNA) fragments.

 User Instructions

The programmable temperature gradient allows you to identify the optimal annealing temperature in a single PCR experiment by letting you test eight incubation temperatures at once. The 1000-series gradient feature uses dynamic ramping, which means the temperature gradient forms during ramping and all wells come to the designated temperature at the same time. As a result, the incubation period is the same for all samples, which is crucial for transfer of gradient to nongradient protocols. The O-ring hermetic seal extends the life of the heating and cooling elements. Condensation from constant heating and cooling can degrade thermal electric modules (TEs), causing them to fail. Bio-Rad’s 1000-series cyclers include a patented* O-ring seal (red in illustration) that forms an airtight barrier around the TEs, preventing exposure to condensation. This substantially lengthens the life of TEs and guarantees outstanding thermal performance.
